• Stable pool generation by electroporation.
• Screening for stable clones using Clone Pix2 (CP2) followed by Limiting DIlution (LD) for monoclone generation.
• Executed expansion, maintenance, cell bank preparation, Production experiments (Fed batch and Batch) and phenotypic Stability studies for Transfected pools, CP2 and LD clones .
• Conducted various Process, media and feed optimization experiment for enhancing Product profile.

Cell and Gene therapy: Development of CART

• Thorough Literature review for Lentivirus (LV) and CAR T process development to design and set up small scale experiments for POC study.
• Optimization of Transient transfection in HEK Adherent cell line for Lentivirus production.
• Performed Total and functional Viral titer analysis for LV.
• Carried out Lentivirus purification by Affinity Chromatography.

Quality Management system

• Preparation of SOP, Clone transfer reports, Tech transfer documents.
• Ensured that all changes made were documented correctly as per QMS and communicated effectively throughout the team.

• Execute transient expression of proteins at Shake flask level using ExpiCHO-S, HEK293, Expi293 in given time lines.
• Expression optimization for transiently expressed proteins.
• Maintenance and cryopreservation of cell lines.
• Analysis of expressed proteins through small scale purification, SDS-PAGE, western blot analysis and Protein A HPLC.
• Contemporaneous documentation of project data and report preparation.
• Perform plasmid isolation for transient expression.

• Supported in downstream processing of Insulin using Preparative Chromatography.
• Performed Analysis of in-process and purified sample using Reverse phase chromatography.
